The generation and analysis of genetically modified mice, including transgenic and knockout animals, is an essential technology for cancer research. The Genetically Modified Mouse Models Shared Resource (GMMMSR) makes these powerful technologies available to all Cancer Center members and others.
Services include:
CRISPR/Cas9 Genome Editing Services. GMMMSR provides comprehensive CRISPR/Cas9 genome engineering in mice, including INDEL generation, targeted deletions, point mutation or epitope-tag knock-ins, and large-fragment knock-ins via the Easi-CRISPR platform. Complimentary consultation is available for CRISPR design. Clients design and supply all CRISPR reagents; GMMMSR performs microinjection of CRISPR components into fertilized mouse oocytes. This service includes a performance guarantee: production of either ten live pups or two CRISPR-positive founders, whichever is achieved first. For projects targeting genes in which the null allele is associated with embryonic lethality, GMMMSR follows a modified injection strategy to maximize recovery of viable edited founders. In the initial injection session, CRISPR RNP complexes will be delivered at 50% of the standard concentration, while the donor template will be delivered at full strength. If a second injection round is required, CRISPR RNP will be reduced to 10% of the standard concentration, with the donor template again delivered at full strength. Completion of these two injection attempts constitutes fulfillment of the CRISPR service request.
